My list at this minute (and it's very liable to change):
The Big Lebowski: One line will suffice to show how awesomely funny I find this movie - unfortunately, there's so many good ones that choosing only one of them is next to impossible.
Shaun of the Dead: Perfect movie for anyone who's ever watched a rom com and thought 'what this needs is more zombies'. It's utterly hilarious.

Some Like It Hot Hilarious all the way through, with the best ending line of all time.
The Forty Year Old Virgin A new entry but a good one. I laughed all the way through.

I concur with the Big Lebowski. I find Office Space hilarious. Best in Show always is good for some laughs.

"Hello, Pickle!" I love SotD. It's one of the few zombie movies I can watch.
I have to give it up for Monty Python and the Holy Grail. That is still so frickin' hilarious! The Holy Hand Grenade scene is one of my two all-time favorite comic scenes in a movie.

It's just so obvious, but Airplane! This is the absolute zenith of the ZAZ chuck-every-gag-you-can-think-of-at-the-screen school of filmmaking. The Naked Gun (first one) comes pretty damn close too. Love final driver's ed instruction.
Wayne's World - maybe it's just the age when I saw it, but I still love this movie to death and annoy the crap out of people quoting it. It's like a pair of underwear...
Van Helsing - Oh, you mean intentionally funny movies??

i must have a weird sense of humor because I didnt find any of the movies mentioned so far as very funny except SOME LIKE IT HOT.
I like MRS DOUBTFIRE. First time I ever saw it, I couldn't breath.

This is Spinal Tap. I laughed almost without ceasing through the entire film.
The Birdcage, with Robin Williams and Nathan Lane. Same here. This is one of my very favorite movies, and if I'm feeling blue, I pop it in and my mood lifts from the first refrains of "We are Family." Everyone in the entire cast (with the possible exception of the son) is brilliantly cast and I think Nathan Lane deserves and Oscar for this film. Just a complete gem of a movie.
